- Cells were placed in wells cut in a thin layer of 0.8% agar in aplastic dish. Folic acid was placed in another well. The cells wereallowed to sense the gradient for 4–5 hours before imagingevery 10 seconds for 5 min. Data are the averages +/− thestandard error of the mean. * indicates significant difference fromwild type. Differences were checked for significance using a 2tailed Student’s t-test p<0.05. Data are the compilationsof 40–50 cells tracked over plastic under agar. The distancemigrated is in 5 minutes. Terms are defined in the legend to Figure1 and in methods. AmpA overexpressingcells move significantly farther and faster than wild type, buttheir directionality and chemotactic index are unchanged.AmpA knockout cells move in a statistically similarmanner to wild type and not faster as they did on the glasssubstrate.
