Smoothened Agonist Activates Hh Signaling and Improves Phenotype of Leptin-deficient HSCs. Primary HSCs isolated from either WT or ob/ob mice were cultured in serum-containing medium for 4 days to induce activation and then treated with SAG (0.3 μM, a Smo agonist that activates Hh signaling) or DMSO for 3 additional days. RNA was isolated, and changes in (a) Hh target genes (Gli1 and Gli2), (b) MF (αSMA and Col1α1) and quiescence (PPARγ) gene expression were evaluated by qRT-PCR. Results were normalized to WT HSCs treated with DMSO.
