Cytosolic proteins from each tissue were applied to a anionic exchange (DEAE-sephadex) column and the flow through (unbound protein) was applied to a cationic exchange (CM-sepharose) column. The bound proteins were then eluted with KCl gradient. Fractions were collected and assayed using tritium labeled dT (filled circles) and dC (open circles) as substrate. TK2 and dCK bind to DEAE-Sephadex column and TK2 eluted at low KCl concentration and dCK eluted at higher KCl concentration.
